Optimization of Amylase Production in Three Fungal Species

Aim: Amylase is an important enzyme that employed in starch processing industries, used the hydrolysis of polysaccharides such as into simple sugar constituents. In this study, we investigated abilities several isolated amylolytic soil fungi to produce amylase.
 Materials and Methods: Soil samples collected from botanical garden, Department Plant Science Biotechnology, University Jos was serially diluted screened for presence amylase producing fungi. Optimization studies performed across different parameters; Incubation period (7 days), temperatures (25-60°C), pH (5-9), concentration (0.2-2%), carbon source (sucrose, maltose, lactose).
 Results: A total 15 isolates belonging 7 genera were isolated. analyzed their ecological parameters. The plate assay showed three species T. viride (62mm), P. citrinum (50.25mm), A. niger (67mm) had largest zones clearance highest activity thus selected further studies. For submerged fermentation, optimum observed at 24 hours incubation all (7.92 IU/ml), (5.04 (7.00 IU/ml). Maximum temperature 45°C (17.10 IU/ml) viride, 50°C (33.60 citrinum, (14.30 niger. maximum 9 (20.40 11 (18.50 (25.80 recorded 15.40 IU/ml 13.20 respectively when sucrose a while 7.28 maltose used. Starch 2% 16.52 IU/ml, 15.4 14.00 respectively.
 Conclusion: Trichoderma Penicillium Aspergillus potential which useful biodegradation biological wastes.
